چکیده

The nuclear and microsomal RNA of three minimal de viation hepatomas and 10-day-old neonatal rat liver were compared by standard DNA-RNA competitive hybridiza tion techniques. nRNA from 5123C hepatoma and 10-day neonatal liver lack sequences present in adult liver nuclear RNA. These missing sequences appear to be identical in part. nRNA from the slowly growing Morris 9618 hepa toma could not be distinguished from adult liver nRNA. The microsomal RNA from this tumor and the 7800 hepa toma both showed sequences to be missing which were pres ent in adult liver microsomal RNA, whereas microsomal RNA from the Morris 5123C hepatoma could not be dis tinguished from adult liver microsomal RNA in relation to their competitive efficiency for liver nuclear RNA. The significance of these findings in relation to transport of mRNA from nucleus to cytoplasm and in cytoplasmic mRNA stabilization is considered.

Competitive DNA-RNA hybridization of microsomal and nuclear RNA in normal tissues of the rat.

Purified nuclear and microsomal RNA from rat brain, spleen, kidney, and liver were compared by means of com petitive DNA-RNA hybridization. In the experiments de scribed in this report a close similarity in the sequences de tected by this technique is noted in the nuclear RNA of kidney, brain, and liver, while a portion of these sequences appear to be missing from the nuclear RNA of spleen. Distribution and specificity of sequences in polyadenylated nuclear RNA of normal, regenerating, and neoplastic liver.

We determined the homology between nuclear RNA of normal liver and that of regenerating and neoplastic livers. Because of the very low representation of the rare class of sequences (1 copy per cell or less) in liver nuclear RNA, very high RNA/cDNA ratios (as high as 1 X 10’) were generally necessary to obtain maximal levels of hybridization. Complementary DNA prepared from 16-h regenerating liv...

Primary effect of dimethylaminoazobenzene on transcription of the unique rat liver DNA.

At present there is convincing evidence of alteration of the RNA population in tumors [l-6] . Study of the alterations in RNA population after carcinogen treatment before the appearance of the tumor is of great importance for understanding carcinogenesis mechanisms There are many papers dealing with the effect of different carcinogens on the RNA synthesis, both inhibitory and stimulating [7]. A...

Analysis of loss of nuclear RNA in azo dye-induced hepatoma by DNA-RNA competitive hybridization.

DNA-RNA hybridization studies, using nuclear RNA's (nRNA's) labeled in vivo and in vitro with high specific radioactivities, were performed to compare the nRNA populations of normal rat liver, livers treated with 3'-methyl-4-dimethylaminoazobenzene (3'-Me-DAB), and 3'-Me-DAB-induced hepatomas. The study with normal liver nRNA labeled by i.p. injection of [3H]orotic acid indicated that the nucle...
